What Is Retatrutide?

Retatrutide is an peptide first "triple agonist" of its kind. Retatrutide activates three hormone receptors: GLP-1, GIP, and glucagon. It is administered as a once-weekly injection.

How Retatrutide Works

GLP-1 and GIP activity reduce appetite and improve blood sugar control, while glucagon receptor activity increases energy expenditure and fat burning in the liver. This three-way mechanism distinguishes retatrutide from semaglutide (GLP-1 only) and tirzepatide (GLP-1 and GIP).

Clinical Research on Retatrutide

Retatrutide is being studied in the Phase 3 TRIUMPH program for obesity, as well as for type 2 diabetes, fatty liver disease, and cardiovascular and kidney outcomes. In the TRIUMPH-1 trial, participants on the highest dose lost an average of 28.3% of their body weight over 80 weeks.

Regulatory Status

Retatrutide is not yet approved by the FDA or other regulators and is available only in clinical trials.

Safety Considerations

The most commonly reported side effects in trials were gastrointestinal, including nausea, vomiting, and diarrhea.
